朱毅敏,1996年6月获天津医科大学临床医学专业硕士学位;1991年8月-1999年9月,任职天津第一中心医院神经内科,住院医师;1999年9月-2000年6月,天津第一中心医院神经内科,主治医师,总住院医师;2000年7月-2004年7月就读于新加坡国立大学医学院生物化学系,2005 年9 月获博士学位;2003年9月-2005年11月,为新加坡国立大学医学院生物化学系研究员;2005年11月-2008年1月,美国加州大学圣巴巴拉分校化学工程系,博士后研究员。现为我所研究员。主要致力于肿瘤靶向性药物的研究。
目前主要研究领域: 1. 功能性多肽筛选技术; 2. 纳米靶向药物载体构建; 3. 纳米材料生物学效应。
本项目组的主要研究领域包括: 实验室目前的研究重点在于针对肿瘤特异性分子或者特定类型的肿瘤细胞筛选靶向多肽。特异性靶向多肽的筛选主要运用了细菌多肽展示技术以及流式细胞技术。细菌多肽展示是一种蛋白质工程技术,可以在体外研究多肽与肿瘤特异性分子或者特定类型的肿瘤细胞的相互作用。通过基因工程技术,把编码不同的多肽序列的核酸序列转入到细菌中,使多肽连接到细菌表面的骨架蛋白上,形成一个融合蛋白一起表达出来。而流式细胞仪作为一种高通量的检测和分选的仪器,在很短时间里能够完成对目标样品的分析和收集。把流式细胞术和细菌多肽展示技术结合在一起,进行荧光激活细胞分选。把靶细胞或靶分子与表达随机多肽序列的细菌库(表达绿色荧光蛋白)进行孵育,通过流式细胞仪进行检测,并分选出带有荧光的细胞或分子,通过多轮的分选与富集,最后得到能够与靶细胞或靶分子特异性结合的多肽序列。 利用筛选得到的多肽进行更深入的研究与商业化产品的开发,主要分为以下四点:1)基于靶向多肽的微流控芯片检测循环肿瘤细胞。2)肿瘤干细胞特异性结合多肽的筛选及应用。3)蛋白酶底物序列的筛选确定与应用。4) 将纳米技术与多肽相结合,设计、构建具有特异性靶向作用的纳米药物以及此纳米药物的生物效应检测。
